A model for the study of skeletal anomalies in rat fetuses BABT
Effting,Cristiane; Paula,Daniel Jesus de; Nunes Junior,Guilhermino Pereira.
The aim of this study was to validate a model of skeletal anomalies in rat fetuses by the administration of ketoconazole (80 mg/kg) to pregnant rats during organogenesis. Bones of the head, trunk and anterior and posterior limbs were examined for detection of anomalies. Statistical differences regarding the number of fetuses and postimplantation resorptions, and fetal and placental weight were significant. The frequency of skeletal anomalies in the head, trunk, and anterior and posterior limbs in the ketoconazole-treated group were also significant when compared to the control group. It could be concluded that the model suggested was valid for study of skeletal anomalies and abnormal bones development in rat fetuse, in spite of the loss of fetuses due to...

Biosynthesis and metabolism of steroid hormones by human adrenal carcinomas BJMBR
Brown,J.W.; Fishman,L.M..
Over a 15-year period, our university-based laboratory obtained 125 adrenal tumors, of which 15 (12%) were adrenal cortical carcinomas. Of these, 6 (40% of the carcinomas) occurred in patients with clear clinical manifestations of steroid hormone excess. Adrenal cortical carcinoma cells derived from the surgically resected tumors in 4 of these patients were isolated and established in primary culture. Radiotracer steroid interconversion studies were carried out with these cultures and also on mitochondria isolated from homogenized tissues. Large tumors had the lowest steroidogenic activities per weight, whereas small tumors had more moderately depressed enzyme activities relative to cells from normal glands. In incubations with pregnenolone as substrate, 1...

Development of microbial and chemical MST tools to identify the origin of the faecal pollution in bathing and shellfish harvesting waters in France ArchiMer
Gourmelon, Michele; Caprais, Marie-paule; Mieszkin, Sophie; Marti, Romain; Wery, Nathalie; Jarde, E.; Derrien, Marc; Jadas-hecart, A.; Communal, P. Y.; Jaffrezic, A.; Pourcher, A. M..
The microbiological quality of coastal or river waters can be affected by faecal pollution from human or animal sources. An efficient MST (Microbial Source Tracking) toolbox consisting of several host-specific markers would therefore be valuable for identifying the origin of the faecal pollution in the environment and thus for effective resource management and remediation. In this multidisciplinary study, after having tested some MST markers on faecal samples, we compared a selection of 17 parameters corresponding to chemical (steroid ratios, caffeine, and synthetic compounds), bacterial (host-specific Bacteroidales, Lactobacillus amylovorus and Bifidobacterium adolescentis) and viral (genotypes I-IV of F-specific bacteriophages, FRNAPH) markers on...

Beltrame,Flavio Luis; Pessini,Greisiele Lorena; Doro,Dani Luce; Dias Filho,Benedito Prado; Bazotte,Roberto Barbosa; Cortez,Diógenes Aparício Garcia.
In this work we investigated the antidiabetic and antibacterial effect of Cissus sicyoides (CS) from Brazil. Diabetic rats that received water (A group) or extracts from the aerial parts of the plant (Cs group) during four weeks were employed. After this period, serum levels of glucose, cholesterol and triglycerides were measured. Glycemia was not affected by treatment with CS. However, there was an increased cholesterol and triglyceride level in Cs group. In addition, bioassay-guided fractionation of methanolic extract from aerial parts of CS was performed for isolation of antibacterial compounds.beta-Sitosterol and sitosterol-beta-D-glucopyranoside isolated showed antibacterial activity against Bacillus subtilis with minimal inhibitory concentrations...

Expression of mdr isoforms in mice during estrous cycle and under hormone stimulation Genet. Mol. Biol.
Schiengold,Marion; Schwantes,Lavínia; Ribeiro,Maria F.; Lothhammer,Nívia; Gonzalez,Tatiana P.; Chies,Jose Artur Bogo; Nardi,Nance B..
The multidrug resistance (MDR) phenotype is associated with the expression of P-glycoprotein (Pgp), coded by the multigenic mdr family. Mice present the isoforms mdr1 and mdr3, which are responsible for multidrug resistance, and mdr2, that is involved in the transport of phospholipids. mdr1 expression has more recently been associated also with the secretion of steroid hormones. This work presents an RT-PCR analysis of the expression of mdr isoforms, in several organs of mice during different phases of the estrous cycle. Additionally, females were ovariectomized, submitted to different hormone treatments, and their uterus was analyzed for the expression of mdr isoforms. The results show that in the adrenal gland and ovaries mdr1 is the main isoform during...

Fatal Strongyloides stercoralis hyper-infection in a patient with multiple myeloma BJID
Yassin,Mohamed A; El Omri,Halima; Al-Hijji,Ibrahim; Taha,Ruba; Hassan,Reham; Aboudi,Kamal Al; El-Ayoubi,Hanadi.
Strongyloides stercoralis (S.S.) is a human intestinal parasite, which may lead to complicated strongyloidiasis. We report a case of disseminated strongyloidiasis following the treatment of myeloma. The patient developed skin lesions, respiratory distress, aseptic meningitis and bacterial and fungal sepsis. The diagnosis of strongyloidiasis was established through endotracheal tube secretions. Despite the treatment with Ivermectin and Albendazole, the outcome was fatal. The value of screening for strongyloidiasis is unclear but may be of benefit in patients with hematological malignancies from high endemic areas

Gonadal function arrest in fillies after birth, as reflected by steroid serum concentrations Braz. J. Vet. Res. Anim. Sci.
NOGUEIRA,Guilherme de Paula; BARNABE,Renato Campanarut; MOREIRA,Alankardson Ferreira; VERRESCHI,Ieda Terezinha do Nascimento.
Sexual maturation involves restraint of gonadal activity from birth to puberty, while somatic development continues. Fetal gonadal steroidogenesis is very important for the maintenance of pregnancy and foaling in mares. The present study was conducted to evaluate gonadal steroid serum levels in foals after birth. Five fillies born at Equília Stud Farm, Avaré, SP, Brazil, were studied. Blood samples were collected daily in the morning during the first week of life. Progesterone serum levels were measured by a commercial radioimmunoassay (RIA) kit and estradiol serum levels with a sensitized non-commercial RIA. At birth, both progesterone and estradiol serum levels were high (13.46 ± 5.5 nmol/l and 7.95 ± 1.5 nmol/l) and decreased to undetectable levels at...

Liaison de l'oestradiol 17 beta dans les hepatocytes de truite Arc-en-ciel et induction de la vitellogenine et de son ARNm specifique ArchiMer
Maitre, J; Mercier, L; Dolo, L; Valotaire, Y.
In the liver of rainbow trout (Salmo gairdnerii ), vitellogenin (Vg) synthesis is influenced by oestradiol (E sub(2)) which is believed to act through the classical mechanism of steroid hormone action. After binding of the hormone to a soluble specific receptor protein, the oestradiol-receptor complex is translocated to the nucleus, where it interacts with DNA and modulates expression of Vg genes, leading to increased synthesis of specific mRNA and Vg. The authors show here: (i) the presence of specific oestrogen receptors in the cytosol of the male trout liver. (ii) The male liver, offering, an ideal experimental control of "zero" background, the authors followed - in the liver of male trouts - the kinetics of induction of Vg mRNA by hybridization with Vg...

Maturation and spawning of tropical and subtropical marine finfish by environmental manipulations ArchiMer
Roberts Jr, D.
Circannual, diel, and lunal periodic mechanisms in naturally reproducing stocks of tropical and subtropical fishes are reviewed. Manipulations of biological clocks, used to control reproduction in captive broodstocks, are detailed. Experiments to induce controlled maturation and spawning in fishes either test efficacy of changing environmental conditions (regimes), or constant environmental conditions (controlled). Data have been generated in experimental/scientific studies, as well as applied research. Effects of controlled environmental regimes on serum steroids, gametogenesis, spawning, fry performance and egg quality are summarized for selected members of Centropomidae, Serranidae, and Sciaenidae.

Modulation of spermatozoon acrosome reaction Biol. Res.
Vigil,Pilar; Orellana,Renán F; Cortés,Manuel E.
Spermatozoon acrosome reaction is an exocytotic event of the utmost importance for the development of mammalian fertilisation. Current evidence shows that the triggering of the acrosome reaction (AR) could be regulated by the action of diverse compounds, namely, metabolites, neurotransmitters and hormones. The aim of the present review is to describe the modulating effects of several compounds that have been classified as inductors or inhibitors of acrosome reaction. Among AR inductors, it is necessary to mention progesterone, angiotensin II, atrial natriuretic peptide, cathecolamines, insulin, leptin, relaxin and other hormones. Regarding the inhibitors, oestradiol and epidermal growth factor are among the substances that retard AR. It is worth mentioning...

Osteoarthritis in horses - Part 2: a review of the intra-articular use of corticosteroids as a method of treatment BABT
Souza,Maria Verônica de.
ABSTRACT: The problem considered in this review is related to the frequent use of corticosteroids (COs) (i.e. steroids) in the practice of equine medicine and surgery as a therapeutic strategy in the treatment of degenerative joint disease, commonly known as osteoarthritis (OA). This disease is one of the most common among the equine musculoskeletal diseases, and is clinically characterized by pain, lameness, joint effusion at the initial stage, which usually reduces with the progression of the disease, and reduced physical performance. Although steroids are considered excellent drugs in the control of clinical signs resulting from osteoarthritis, they also influence cellular activity through activation of various signaling mechanisms. However, they can...

P-glycoprotein-mediated resistance to chemotherapy in cancer cells: using recombinant cytosolic domains to establish structure-function relationships BJMBR
Di Pietro,A.; Dayan,G.; Conseil,G.; Steinfels,E.; Krell,T.; Trompier,D.; Baubichon-Cortay,H.; Jault,J.-M..
Resistance to chemotherapy in cancer cells is mainly mediated by overexpression of P-glycoprotein (Pgp), a plasma membrane ATP-binding cassette (ABC) transporter which extrudes cytotoxic drugs at the expense of ATP hydrolysis. Pgp consists of two homologous halves each containing a transmembrane domain and a cytosolic nucleotide-binding domain (NBD) which contains two consensus Walker motifs, A and B, involved in ATP binding and hydrolysis. The protein also contains an S signature characteristic of ABC transporters. The molecular mechanism of Pgp-mediated drug transport is not known. Since the transporter has an extraordinarily broad substrate specificity, its cellular function has been described as a "hydrophobic vacuum cleaner". The limited knowledge...

Role of steroid hormones and external factors in the control of pituitary gonadotropic activity of female European eel (Anguilla anguilla ). ArchiMer
Leloup, H; Fontaine, Y; Dufour, S; Querat, B.
Gonadotropic activity is relatively low in the female freshwater (FW) eel (A. anguilla ): pituitary GTH was about 15 ng equivalent cGTH/mg fresh tissue. Plasma GTH was always less than 1 ng equivalent cGTH/ml and often undetectable (< 0.2 ng equivalent cGTH/ml). Ovarian steroidogenesis in FW fish is similarly low; plasma concentration of both free testosterone (T) and free estradiol (E sub(2)) were less than 0.3 ng/ml as measured by RIA. Treatment with E sub(2) in FW fish caused a great increase in pituitary GTH, up to 1000 times control values, without changing plasma GTH. Stimulation of gonadotropic activity and ovarian development occur during the eel's migration to the Sargasso Sea. Adaptation to seawater (SW) caused a great increase of plasma E...

Triterpenes of leaves from Piranhea trifoliata (Picrodendraceae) Acta Amazonica
JEFFREYS,Manoel Feitosa; NUNEZ,Cecilia Veronica.
ABSTRACT The Amazon forest is rich in plant species diversity, among them,Piranhea trifoliata stands out, which is popularly known as piranheira, because their fruits are eaten by fish. Their barks are used as bath composition on uterus inflammation and as tea in malaria treatment. This study aimed to fractionate the dichloromethane and dichloromethane phase from methanolic extract of leaves of Piranhea trifoliata. The leaves were dried, grounded and extracted with dichloromethane, methanol and water. The methanol extract was partitioned with dichloromethane and ethyl acetate. The chromatographic fractionation yielded six pentacyclic triterpenoids: friedelan-3-one, 28-hydroxy-friedelan-3-one, 30-hydroxy-friedelan-3-one, lupeol, α- and β-amyrin mixture,...

Worker honeybee hemolymph lipid composition and synodic lunar cycle periodicities BJMBR
The aim of the present investigation was to extend a previous study, showing a correlation of the variations of hemolymph carbohydrates with synodic lunar-like cycle and its circaseptan harmonics to worker honeybee hemolymph lipids. Hemolymph lipid concentrations of emerging worker imagos were analyzed in terms of one ideal synodic lunar cycle and processed by the cosinor method testing the null hypothesis versus the presence of 29.5-, 14.8- or 7.4-day periods in the data. A rhythmicity statistically compatible with a 29.5-day rhythm was observed for triacylglycerols and steroids as well as for body weight. A circadiseptan rhythm was determined for 1,3 diacylglycerols, while fatty acids and phospholipids exhibited a circaseptan rhythm. An agreement of...
